If I understand correctly you want to separate the coloured blobs from their respective backgrounds.
Forget the blend modes (nod to Walter “transfer modes”).
The best way to do this is to create a matte based on the colour values in the layer.
One way is to apply a linear colour key to the layer and key out the white. This should reveal the blobs alone.

If you precomp the Shatter you can use Time Remapping to reverse, stop and play forward again in any way you’d like.

Text animation is controlled by animators.
I assumed you had some understanding of the software.

You’ll need to animate the Character Offset using 2 separate animators, one for the last digit and one for the penultimate digit.
Restrict the range of each animator to a single digit. The penultimate one should only animate when the the last digit turns from 9 to 0.

This looks like a problem with your video card driver, not After Effects.
Update your driver and make sure you are running the current release for the version of After Effects you have.

I’m not sure why you’re receiving that error but mp3 files should be avoided in After Effects. Convert the file to an aif or wav.
MP3 is a compressed format and will only compress further in AE resulting in audio artifacts.
