I would suggest you to go with what Joseph has suggested.Using a black solid with wiggle expression on opacity.
You can also control the amount of wiggle using a simple expression.

There are many ways to do this:
You can use different lights to interact with your scene. Your footage should be 3D (to get affected by light) and it should look natural so you have to work hard on this point.
For nightclubs lighting effect, Optical Flares and Knoll Light factory will help.

I agree with Joseph. It might be shot with a green screen and then keying the character or rotoscoping the character if the bg is real.

The dotted backward(not forward)slash means that it is in Draft Quality.
Change it to Best Quality(Forward slash) as it was earlier.
Also check that you are rendering with Best Setting and Full resolution.

Sorry but i didn’t animate the stroke.

If you Check “Force alphabetical order” option while importing the sequence, Ae will not show any error.

You can use any color for BG but make sure it does not appear on the subject.
