Without seeing the image you are talking about its a crap shoot but here goes…
In AE on one layer you could draw a matte around the clouds you want the lightning (you spelled lighting but Im assuming you wanted lightning)
so the cloud is isolated.On the next layer (below the first) copy the same clip.
Then sandwich in between these two layers another plain layer. On this one apply the AE flare effect. Animate the opacity and placementand blend it to taste.
Just one of many ways. And alternate would be to assign a glow to the middle layer which now is a dup of the other two . Animate the glow . Mask it if necessary.

FWIW i think this might be a Red related issue. Ive done lots of HD editing using the default MPEG I frame for Previews (rendering on the timeline) and never saw what you are describing.

Well when all else fails. Create a brand new project. Then import your current project into it. See if that helps.
Like Dennis said, check your media raid to see how full it is (Im assuming you have a raid if you are trying to play back 5K 😀 )
Is your graphics card running CUDA? I know its ATI so Im not sure its supported.

Does the frame rate of your footage or comp in AE match the footage that was tracked in PF Tracker?

Try this..
1. Delete Preferences (boot up PrP holding down Option (Alt) and Shift keys .
2. Delete Media Cache files
3. Delete Preview files
Find out where the folders for step 2 and 3 are. Close Premiere. Delete files inside both these folders.
