It depends on what you want to do. For me personally, I spend most of my time in After Effects (AE). I do know Photoshop and I think it is important to at least know the basics. But for the stuff I create most of it is done In AE.

I would use the shape tool to create it and add the glow filter or play with blurring it and adding a blend mode.

The link doesn’t work. Red Giant has a bunch of free pre built particles that you can download. Check out: https://people.redgiant.com/ However, they just Announced Red Giant Universe which will have the free presets and much more. I would check out Red Giant Universe.

There are many ways to approach it. Depending on the complexity of the animation will depend on how complex the blinking needs to be. I just cut this spot and the characters are very simple:
Because of that I had the eyes on a separate layer and just created a mask in the middle of the eyes. Mask opacity set to zero, next frame mask opacity set to 100. You see half the eyes at that point. Then mask scale changed to cut eyes out completely. Next frame set to old mask scale. Next frame zero opacity.
