Most likely cause is a codec that is not supported. What are the properties of the clip? Does it play fine in the Source window and not in the Program window? What is the source of the clip?

VirtualDub is a freeware converter…google it.

Premiere Pro cant help much with Mpeg 2 files.
Option #1 Main Concept’s Mpeg plug-in for Premiere ($400)
Option #2 Plug your camcorder into a analog/digital converter and capture as DV into Premiere ($160 and up).
Option #3 Premiere Elements 3.0 ($99) -
A drive with large files might not defrag completely but this should not cause your issues. Do you have only one drive? Suggest updating video and sound card drivers.

Try updating QT player or uninstall it and reinstall.

To check a drives health, right click on My Computer and choose Manage. Expand Storage in the left pane and select Disk Management…what is the Status of the drive? -
For a test I exported a DV NTSC.mov file from QT Pro…2 minutes, 427 MB. Pro does bog way down during conforming audio which took about 1 minute, but then it played fine on the timeline (P4 2.8 HT). Qt player plays the file fine.
Check the properties of your file in QT…mine shows Apple DV as the format. -
“Rendering” preview files should be only necessary where you need a high quality preview of part of your timeline that your PC cant play smoothly, like a graphics/motion or slow motion sections. I seldom render much at all.
Also forget rendering nested sequences. I seldom do anything with nested sequences other line them up and then export to Mpeg 2 for DVD.
Preview files are not used when exporting to DVD.
I manually delete preview files as soon as the project is done. -
Create a new empty document big enough for both parts. Drag each part into the new document and align as needed.

Windows Media Video supports URL “flips”. At a point during playback, typically at the end, the URL marker created in Premiere launches their browser to a web page.
To Create a URL link within the video screen requires an Mpeg 4 or Flash approach. Premiere can export .flv files used in Flash authoring, and the H264 Mpeg 4 files used in SMIL (simultaneous multimedia integration language) and other authoring environments.
