It depends on the TIFF. The likely culprits are that the compression (if used) is bad or the file is otherwise corrupt. Could also be the file was saved with a legacy formatting (doubtful). If it won’t open in AI, but it will in PS, it was likely corrupt in some way, but AI isn’t as robust as Photoshop when it comes to raster files anyway.

Honestly, I think you’re better off using something like Indesign to do this. It has many detailed text tools that will really make the job easy. You can always import a PDF version back into Illustrator later on.

So these are just alternating blacks and whites? Shouldn’t take long even doing this manually, but it will take some time to do it if you are new to Illustrator. Basically, you just need to keep drawing. The more you do it, the easier it gets, but you have to put the time in first.

Sounds like you are doing a clipping mask. Arrange your 2 objects (assuming both are vector objects) by positioning first the colored part how you want it, then put the paint daub above it. Select both objects, then go to Object >> Clipping Mask >> Make or Command-7 or CTRL-7.
If they are not both vector objects, use Photoshop to combine them.

Okay if you are dealing with TIFFs or JPEGs, you are better off working in Photoshop. In PS, open your TIFF, make sure you are set to CMYK for your color mode, you may have to convert if not, but that may not work correctly. Anyhow, you can simply isolate the Cyan (C) channel in the channels palette, CMD-click (CTRL-click for PC) the channel to select it, then create a new channel, assign it a spot color, then paste the info to the new channel.

All of the changes you make to the image in RAW are non-destructive; you can always go back to the original and make changes. Further, I find RAW much faster than doing the same actions in PS. Also, if you import an image as a RAW smart object, you get the best of both worlds.
