I am not usually using OpenGl in AE- it tends to mess up things a lot. But I did try to use it with the new Invoigorator Pro- I got the same, OpenGL previews – just fine, but then it would not render using OpenGL. I got the “not supported” message although it is obviously supported in previews. Todd or anyone else…?
Animation codec is rarely going to play smooth on a good system- you need one build to handle uncompressed video. It is normal to get a large file size and it is also normal to have the clip stop or even not to play if the frame size is quite large as in your situation. Take that .mov and convert it using Adobe Media Encoder and see if your compressed file plays ok.

Maybe duplicate the layer and “drill” holes through it, then composite on top of base skin with drop shadow that is a darker skin tone color and use matte choker to open and close the “pores”. That’s the first thing that comes to mind.
Another more fancy option would be to use Free Form and create a displacement map for the pores- you can then animate the displacement in 3d (negative displacement) and use AE camera and lights to make them visible by casting shadows and slowly moving the camera to change perspective…
You can draw a path, animate that for the length of your clip to match the shapes you need (you can even track the movement of the background, apply that to a Null and link the path to it). Then Link Text to follow the path.
Use Adobe Media Encoder to render out your project. You can open a AE project in AME and render to MPEG2 for 1920x1080p 25 fps. This should give you a good quality output.
