You want to make a still image look like it’s video or film? (I interpreted ‘camera rolling’ to mean “camera running”, not “camera rotating”.)

You could add some animated grain.

For simple animations of 3D objects, consider using a Photoshop 3D object layer. Basically, you can import a 3D object into Photoshop and then bring the PSD into After Effects and treat it like a real 3D object. It doesn’t work for really complex things, but it certainly works for the kind of thing that you described.

This page describes how the Puppet tool interacts with paths already on a layer. Basically, if you already have masks on a layer, the Puppet effect is expecting you to click within one of them to define a mesh within that outline.

Do you mean that you want to make your default output module in the render queue one that encodes and exports movies with an alpha channel?

If that’s your question, then see this section of After Effects Help for information about specifying a default output module. Farther down on that page is information about how to make sure that the alpha channel is part of of the output. Be sure to pick a format and codec that can include an alpha channel.
