You could set up a few different brushes to accomplish this in Illustrator.

After the red-eye has turned grey, you can use your circle marquee to select the pupil, then adjust the levels to turn it black. I hope this is what you’re talking about.

Your pixel aspect ratio may be set for horizontal pixels.

I don’t know the keystroke, but I use the history palette to create new document from current state and then flatten the layers with a keystroke I assigned in the Edit>keyboard shortcuts.

GoLive has underline. InDesign has it. Photoshop has it. After Effects has it. etc. But it wasn’t until CS2 that Illustrator got it.

This should help. You can actually skip the step in making the line 99% opaque.

I use Cinematize and it works nice.

Sometimes I’ll pull my files into AE to use some of the shine effects in there, then I export a frame with alpha channels (RGB+) and it works great.
