I’m old school and avoid mixed codec’s in a single project. Just like the big houses, I convert all incoming media to OP1a MXF for video, Wav for audio and PSD for stills, (I have watch folders set up in AME for most of this, and run batch scripts in Photoshop for stills). It’s all native within PPro and renders faster than realtime (google smart rendering in Adobe).
Sometimes the issue is corrupt cache/audio conform files. Therefore, you might want to clean out the data cache and allow new conforms and temp files to be re-created prior to exporting. You might also want to run a disk check. You never know.

+1 with David and master projects.
Been doing this for years. You can go further than that by creating a master folder with sub-folders and included reusable assets as well, (Including Audition and AE projects). Whatever it takes to avoid re-creating stuff and staying organized. Just copy and rename the root folder as the new project name.
However, when you go that route and wish to use Project Manager to reduce/archive finished projects, it will ignore the folder structure and just dump everything into one folder. Memory is cheap. I just copy the entire folder over to an offline drive.

In PPro, check the sequence settings, make sure they match the media. (Alternatively, you could right click on a clip in the bin, choose “new sequence from clip”. Then open and play that sequence).

If you’re using a Windows PC, open the sound panel and make sure that your playback device has it’s switching set to use the application that has focus (exclusive mode):

Anything else sharing the USB buss? Disconnect, move to another port, etc. See if that makes a difference.
Do you have another drive to verify or rule out the port as the issue?

In addition to what Jeff said about not having media assets on the system drive, and using the USB 3.0 port. You have to consider the layout of your system and data flow. It’s not just keeping media files off the system drive, it’s also avoiding any other drives or ports that share resources with the same buss as the system drive. Therefore, if you have more than one USB 3 ports, trying another port might make the difference.
Another thing to consider is audio conforms and cache file locations. Where are those being save to? Same as the project drive, or a temp folder on the system drive?
Think of your PC like a super highway. You want the fastest drives/largest data sets to have the fastest lane to themselves.

It could be a license issue/check, (I have near the same setup as you with a T7610). Occasionally, the application wants to phone home and do a handshake with the license server. In the beginning, I had to lug my workstation home to do this, (including the initial install and to download updates) due to an overzealous security system at work. Which IT remedied later.
There was a time that I had to reinstall CC desktop manager to fix a non-connection issue.
One more thing, do you have admin rights to the workstation? If not, then IT should get involved or logon and run the CC manager to see if it want’s to phone home and reset.
