Switch around the channels using one of the channel effects. So make the R, G, and B all equal alpha.
Or create a white solid. Put the keyed footage one layer above it. Then set the white solid’s track matte to alpha.

no can do in AE. And amazingly this still isn’t offered in AE7 as far as I’m aware. You can put the position frames on a layer that the mask is a part of though.

Yes, add the Particle System to a solid.
I’m at home now so I don’t have those settings in front of me but I’ll tell you what I remember:
Particle: Soft Sphere (or faded sphere)
Turn off all velocity and gravity.
Make the birth and death colors white.
Make the birth size a little bigger than the death size. Set the lifespan to your liking.Zero out any randomness, like random size. Really go and zero almost everything. You just want the particles to exist were they were created. Get it to that then tweak from there.

No, I used the standard AE Glow.
Effects>Stylize>Glow
Actually I placed a gaussian blur (Effects>Blur>Gaussian Blur) between the particle system and the glow as well.

Did you load your CC effects off your installer discs? AE 6.5 has a folder called “Cycore” install these plug-ins. Theres a ton of awesome plugs (used to be called Final Effects). Might as well oad in Color Finesse while you’re at it (in the synthetic Aperture folder).
When it’s installed you’ll see in your effects menu a bunch of plugs with ‘CC’ in front of them. The particle systems will be in:
Effects>Simulation>CC Particle Systems II

I wouldn’t bother w/ the playground. Way over done for this. Use CC Particle Systems II (Free with AE 6.5).
Here I whipped this up with CC Particles Sytems and the standard Glow. With tweaking you can make it much cooler.

Just paste the position keys into the Producer position parameter.

You know if you paste it to a particle emitter it will create a stroke for you. 3D stroke itself is actually evenly spaced particles sprites of white discs along a path. So create it yourself and control the parameters with a particle system.

This is using particles and not strokes. Just use the Basic AE particles to do this. Don’t need particular.

Best thing to do first is search the Cow archives with the keyword “roto”. Make sure to select the last few years. There have been many techniques on how to do it. Also if Barend can point us in the right direction to his FAQ he compiled, there is great info on that.

Ideally if you knew your goal before shooting you would put little tracking dots on your fingertips for the tracker to grab onto. You may have to just do it frame by frame. Hopefully your shot isn’t too long.
