I’ve not heard of such a trick. Indexing and conforming is how PPro helps manage files between it and After Effects as well as, I believe, Audition so it doesn’t need to re-process them at each step.
As far as I’m aware, I think all conforming does is write some XMP data and convert 16 bit audio to 32 for editing purposes. That’s what Adobe’s forums state.

1. Are you given any error message at all or does it just stall?
2. What codec are you using to encode?
3. Is there anything new about this timeline other than length that is not in your other projects? Footage encoded with exotic codecs, heavy After Effects link-ins, etc?
4. When AME stalls, can you look at how much system ram is currently used? It may be maxed out.Those are a few questions to start the troubleshooting process.

If your master output is 25p, your timeline needs to be 25p.
Things to remember
1. Set your timeline to 25p
2. Use two copies of the same file in your media bin (library)
3. If 50p footage is dropped onto a 25p timeline, the footage will conform to play at normal speed
4. For slo-mo, you need to right click your file in the media bin, go to interpret footage and type in 25fps. This is why you’ll want two references in your media bin to the file, so they can be interpreted at full speed and slomo.-Angelo Lorenzo

Lets say if you have a calibrated monitor and an uncalibrated one on the same system, the preview window would look different on each.

I Think I may misunderstand the issue, so let me rephrase it: You color corrected a clip looks amazing, brought it on your laptop to render, brought it back to your color monitoring environment and it looked weird?

Alex, I just think my workflow is adulterated by the fact that a lot of my work is short-notice or have tight deadlines, and clients like to “see things along the way”. I’m sure you’re aware people get weird about seeing stuff in “draft” without at least some sound.
It is what it is, but I’ve been trying to streamline things.

1. What is your color reference monitor?
-How is it connected to your desktop?
-How is it connected to your laptop?
-Are you calibrating with color bars or something like a Spyder 2 (if it’s an additional computer monitor) on both the laptop and desktop?Depending on how it’s connected, it sounds like the graphics system on the “messed up” laptop is probably pumping it through a different LUT… could be a few things going on.

Good recommendation JB… extract always slips my mind. I even avoid Ripple Delete, I have too much going on with my audio and foley tracks to rely on anything but manual shifting. Probably an error in thinking in my own workflow.

Ever try right clicking the gap (command-click on Mac I believe) and selecting Ripple Delete?

Without knowing that, I can only suggest the following:
1. Your program monitor may be zoomed in to 100% instead of “fit”. To correct this underneath your program monitor you will see a drop down menu. Select “fit” or any of the other options until it fits nicely on your screen.2. If it’s a scaling issue you can right-click on your clip in the timeline and deselect “fit to frame size”.

The fact that the video really starts to slow down during the transition makes me think that you may not have the Mercury Playback Engine set to hardware use. The setting is on a project by project basis. Go to Project > Project Settings > General and make sure “Video Rendering and Playback” is set to “Mercury Playback Engine GPU accelerated”.
Also, are you trying playback immediately after importing a new clip, or are you giving it enough time to conform like new media usually needs to by watching the little conform bar in the corner?
