The next thing to try is to close PPro, rename the folder(s) that contain your clips to effectively hide them from PPro. Then when you open the project click on “offline all” and see whether or not the project loads.

Did you try it without any transition at all? If that doesn’t “look right” then I don’t think a transition is going to hide the fact that you are using two clips.
You will also find links to many free tutorials in the PremiereProPedia that will quickly show you how things are done in Premiere Pro.
Ensure that you have the latest ASIO compatible drivers available from the manufacturer’s (RealTek) Web site and that the correct audio driver is selected in PPro.
If the 4.0.1 update does not resolve it then you can try the workaround. Create the Mac equivalent of a Windows Shortcut as described in the thread I referenced.
