Here’s an attempt based on what you described in your last post.
It doesn’t behave at all like the file you supplied, but maybe that’s a good thing? No expressions necessary, just a Null and some parenting.
Maybe you can direct us from here if this isn’t what you’re looking for.

It is really hard to see what you’re trying to accomplish in the file you supplied. Can you describe what this is supposed to be doing in more detail? Is this supposed to behave like a real world wheel in some way?

There are a couple of ways to initiate different RAM Previews:
Numpad 0 – This will start a RAM Preview with the “RAM Preview Options” found in the Preview panel.
Shift + Numpad 0 – This will start a RAM Preview with the “Shift + RAM Preview Options” found in the Preview panel.
You may have checked the box in Shift + RAM Preview Options but are only pressing Numpad 0 or vice versa. Check it out.

Create a new Null, place it at the point you want the spinning layer to spin around and make the Null a parent of the spinning layer. Then you can rotate the Null to get the result you described.

If you have the pixel resolution of the display and the physical aspect ratio you can determine the pixel aspect ratio with some simple arithmetic.
Pixel Aspect Ratio = (Physical Width / Physical Height) / (Pixel Width / Pixel Height)In all likelihood (and in my experience) an LED array will use a square pixel aspect ratio.

My guess is that you want 2 masks on your layer. Keep the current mask and add a new rectangular mask that encompasses the entire layer.
The easiest way to add a layer-sized rectangular mask is to double click the rectangular mask tool in the tool bar with the layer selected (make sure you don’t have your current mask selected though or it will replace the mask shape).
Once you have these 2 masks you’ll want to animate the mask opacity of the rectangular layer sized mask instead of the opacity of your original mask (leave that one at 100%).

You should never need to know dot pitch when working in AE or Premiere. The only things that matter when creating content with AE for a specific display is that display’s pixel resolution and pixel aspect ratio.
You can simply match your composition settings to these values and you should be good to go.

I’m not sure that this is a problem, or at least not the problem you’ve described.
The [0,0] location in an AE comp is the upper left corner, but the values increase moving to the right and down. So in the graph you’ve shown in your post the visible values (that is those between [0,0] and [thisComp.width, thisComp.height]) in the AE comp would be in quadrant I as you desire.
They would be mirrored across the X-axis relative to the AE comp but they should be in quadrant I.

This is what the “Reset Character” and “Reset Paragraph” options I mentioned earlier will do in addition to resetting other possible pitfall parameters. So if it didn’t fix it earlier it probably won’t fix it now 🙁
If you can share a sample problem after effects project we can probably figure it out. You don’t even need to include the footage.

You can upload images directly to the COW and embed or link to them in your posts. I highly recommend doing that in the future for a couple of reasons:
-Most image/file sharing sites are blocked on corporate networks.
-Better (as in less) compression.
The Image Upload button is the third from the right in the small formatting strip above the message box.
