Using AC130 in relay record mode, what is the best way to transfer video from SDHC cards to HD for use in Premiere Pro 5.5

You should have no issues transferring all cards to your hard drive in different folders (one for each card). Then from there just drag into your NLE- spans are generally automatically resolved.

So even if I transfer the files via a SDHC card reader, when I import the files all together into Premiere Pro the overlapping scenes from Card 1 to Card 2 will be stitched together?

If by transfer you mean copy the entire directory folder from each card to your hard drive- yes.
