    I need to be able to create a coloured ‘pop art’ style JPEG ‘on-the-fly’ from an original, regular iPhone JPEG. I’ve attached a Photoshop mock-up of what I’d like to achieve, my question is, without an alpha channel or any kind of prep, can this be done entirely within After Effects?

    combing the threshold effect and tint effect will get you very close….

    add threshold and set the ‘level’ as needed to make a black and white image of the photo.

    then add the tint effect to colorize that, setting ‘map black to’ and ‘map white to’ the colors that you want.

    you could add the curves effect, or other color adjustment prior to the tint effect to give added image control.

    Use Threshold to convert to a purely black-and-white image (no shades of gray), and Tint to color the black areas pink and the white areas orange.

    You can get some different looks using an effect like Channel Combiner before Threshold and choosing the Red, Green or Blue channel.
