GPU will help with some CODECS, notably AVCHD. There is NO GPU acceleration for MPEG or AVI.

I would certainly use an SSD for the boot drive C:, and perhaps use 2 pairs (total of 4) of Western Digital 10,000 RPM VelociRaptor drives in a mirror and stripe configuration. This assumes your motherboard supports hardware RAID and can handle that many SATA connections (I count you would need 6). You could just get by with 2 VelociRaptor drives and not worry about redundancy and instead rely on regular backups to a USB 3 drive. You would most likely need to use a 1000W power supply.
I have used VelociRaptor drives and they do run quite hot, so good case cooling is essential.
My system specs: Intel i7 970, 12GB RAM, ASUS P6T, Vegas Pro 12 (x64), Windows 7 x64 Ultimate, Vegas Production Assistant 1.0, VASST Ultimate S Pro 4.1, Neat Video Pro 2.6
