I am working in Premiere and After Effects on a feature documentary.

    I have 1.5TB of video.

MY QUESTIONS: Do I but the video or the render files on the faster drive. I have both thunderbolt and firewire? I’m thinking of replacing the thunderbolt with a USB 3 drive. Is there a big difference in performance between thunderbolt and USB3?

I will be running the iMac 5k 4.0GHz quad-core Intel Core i7 with 64 gig ram.

I’d put it on the drive with the faster transfer rate, yes.

There are tons of discussions on the web about the speed of different connections. Is yours Thunderbolt 1, 2, or 3?

Thunderbolt 2 is significantly faster than USB 3 and even 3.1. You could test it yourself on your system though to see how it acts for you! Put the same large file on both drives and see how fast it transfers to and from that drive. Then try encoding that file with Adobe Media Encoder from each drive and compare the difference.
