I’m trying to match visuals with sound peaks in the audio.
Can after effects play the sound when manually dragging the slider.
If so how can it be enabled?

No, but you can add markers in the timeline panel during playback by tapping the asterisk key on the numeric keyboard.

Or you could use the “Convert Audio to Keyframes” keyframe assistant: right-click your audio layer, then navigate to Keyframe Assistant > Convert Audio to Keyframes.

Or you could use Trapcode SoundKeys for more control over how the audio is sampled and interpolated.

Oh, sorry! I completely misunderstood you. What you’re describing is called the Current Time Indicator (CTI, also sometimes called the playhead). A slider is an effect control.

I’m glad you found the solution.
