    I would like to be able to animate points on a mask (line, not closed). I have the 3D Stroke effect applied to several splines and want to give them some motion. Short of hand keyframing each spline – which I REALLY want to avoid – is there a more automated/random method to creating movement here.

    Yes, the Script RotAE.jsx, here’s an earlier post on how to apply the wiggle expression to mask vertices. This script was originally for Roto work, but any keyframes that can be generated, the original purpose of the script was to put track Point keyframes onto mask vertices, but manual keyframes or Expressions Converted to Keyframes (your need), will work fine.
