I have been asked to bid on a huge compression project, for DL DVD.

I can estimate file sizes using the various bitrate calculators.
But does anyone know how one might estimate how many hours
such a task might take, given certain computer specs?

    I’d like to bill hourly
and give a ballpark idea of the total.

    Thoughts?

There are a lot of different variable with this. It will largely depend on your computer specs. if you have a good processor and plenty of ram it should be relatively quick. Also the source will affect the speed. i recently did several DVDs from an SD source and encoding the video took about 45 minute – 1 hour per 2 hours of footage. If I were you, I would just run some benchmark tests on the system you’ll be using with footage that is similar to your source to get some ball park estimates. Good Luck!
