What exactly are you trying to accomplish? Other Channel effects (such as Calculations and Set Channels) do work in color depths of greater than 8 bpc.

I have been given a lens flare shot on 2K film and I am trying to give it an alpha channel using the technique Ben Rollason has described in one of his tutorials. Basically Straight to Pre-multiplied/invert.

For that, you can use the Shift Channels effect instead of the Channel Combiner effect. There’s even a link on the “Shift Channels effect” page of After Effects Help that points to a blog post that explains how to do this.
