I have a layered Photoshop file that I am using to animate a character. His arms, legs, etc are each on a separate layer.

If I were to add another layer to the PSD file, will it update in AE? If not, will I a simple replace footage work?

You can use the Edit Original command in After Effects to open a PSD file in Photoshop, make and save changes, and have those changes appear immediately in the movie that refers to the PSD source file. Even if you don’t use Edit Original, you can use the Reload Footage command to have After Effects refresh its layers to use the current version of the PSD file.

It won’t automatically update and add the new layer. You’ll have to do that manually. Just import that layer only and add it to your comp.
