The post of Kevin’s that you refer to was for After Effects CS3. Things have changed to make using these settings much easier.
One thing that makes questions like this very difficult to answer is that optimization depends on each project—actually even on each stage of each project. If there were one perfect set of settings, then these settings would be hard-coded into the software instead of exposed to you as preferences.
That said, the first thing to do is to make sure that you allocate adequate RAM to each After Effects process. Read this post for more details about that. That post also points to the definitive pages about the Memory & Multiprocessing preferences and improving performance in After Effects.

The first thing that leapt out at me:
Don’t use Render Multiple Frames Simultaneously multiprocessing if you’re having trouble completing the render. That feature splits resources among many processes to make rendering faster. If you want to make sure that a heavy composition renders, you want to have the maximum resources dedicated to one process.

I’d use a font that has a dot character and then use the text animation features to animate the font size.

Yes, you can use multiple processor cores with After Effects CS4 on a 64-bit OS. The feature is Render Multiple Frames Simultaneously multiprocessing.
You should also read this post about avoiding some common problems with that feature.
